TheMACSPlexCytokineBasicKitcontainsthesupplementarycomponentsnecessarytoperformaMACSPlexCytokineassay.IthastobecombinedwiththeMACSPlexCytokine12Standard,humanandthedesiredMACSPlexCytokineReagentsKits,humanorwiththeMACSPlexCytokine10Standard,mouseandthedesiredMACSPlexCytokineReagentsKits,mouse,respectively.Donotmixhumanandmousespecificitiesinoneexperiment.MACSPlexCytokineReagentsKitsaredesignedfordetectionofoneormaximaluptosevenanalytesofchoiceinasinglesample.
MACSPlexCaptureBeadswithinthesingleMACSPlexCytokineReagentKitscontainfluorescentlylabeledbeadpopulations,coatedwithaspecificantibodyreactingwithoneoftheanalyteswithinthesample.MACSPlexCaptureBeadsfromdifferentreagentskitsdisplaydifferentfluorescenceintensities.Samplescontainingunknownlevelsofanalytesareincubatedwiththeantibody-coatedMACSPlexCaptureBeads,andanalytesbindtothespecificantibody.EachdetectionreagentcontainsAPC-conjugatedantibodiesspecificforoneanalyteandisalsoadded.Consequently,sandwichcomplexesareformedbetweentheMACSPlexCaptureBead,theanalyte,andthedetectionreagent.ThesecomplexescanbeanalyzedbasedonthefluorescencecharacteristicsofboththeMACSPlexCaptureBeadandthedetectionreagent.StandardsofknownquantitiesofgivenanalytesareprovidedwiththeMACSPlexCytokineStandardsandareusedforthequantificationoftheanalyteswithintheunknownsamples.
TheanalysisofMACSPlexCustomCytokineAssaysrequireaflowcytometerwithablue(e.g.488nm)andared(e.g.635nm)laser,whicharecapableofdetectingFITC,PE,andAPC.IncombinationwiththeExpressModesoftheMACSQuant®AnalyzerstheMACSPlexCustomCytokineAssaysareoptimizedforautomatedmeasurementofthebeads.Theysimplifyflowcytometricanalysisviapredefinedexperimentsettingsaswellasacquisitionandanalysistemplates.Theyapplyanautomatedgatingstrategytopopulationsofinterestthatwillbeautomaticallyadjustedforeachdatafileindividuallytoachieveoptimalresults.
